Studies on the Thidiazuron Treatment of Anther Culture in Paeonia albiflora



Abstract
These studies were carried out to investigate the effects of Thidiazuron(TDZ) on the microspore-originated embryo and callus formation, pollen dimorphism and low temperature pretreatment of floral bud on the emergence of embryogenic S pollen in vivo and in vitro, the origin and development of pollen embryos and calli, organ differentiation from induced embryos and calli.
The anthers of Paeonia albiflora were cultured in Murashige & Skoog¢¥s basal medium supplemented with ¥á-Naphthalenacetic acid (NAA), 6-Benzylaminopurine (BA) and Thidiazuron(TDZ).
The results obtained were summarized as follows
The appearettce rate of S pollen was 28.7% in vivo, while that of S pollen by low temperature pretreatment at 5¡É for 10 days was increased by 53.2%.
The apparence rate of S pollen was 71.9% by low temperature pretreatment for 10 days at 5¡É in 40 days¢¥ cultures.
Embryo was observed by division of vegetative nucleus.
The induction of callus was the best in the medium with 0.2§·/L NAA 0.5§·/L BA and 0.4§·/L TDZ at control and 0.2§·/L NAA and 0.4§·/L TDZ for 10 days at low temperature pretreatment, 5¡É.
The induction of embryo was the best in the medium supplemented with 0.2§·/L NAA and 0.0002§·/L TDZ for 10 days at low temperature pretreatment, 5¡É.
The induction of shoot and root on the redifferentiation medium was the best in the medium containing 0.5§·/L BA or 0.002§·/L TDZ. The formation of secondary embryo was induced in the medium with 0.002§·/L TDZ.
